UniProt ID: | P43431 (α subunit), P43432 (β subunit) |
Formulation: | Lyophilized from a sterile filtered solution, pH 7.5, containing 10mM sodium phosphate, 50mM sodium chloride. |
Purity: | ≥95% (Reducing and Non-reducing SDS-PAGE) |
Endotoxin Content: | ≤1 EU/μg protein measured by kinetic LAL analysis. |
Appearance: | White lyophilized (freeze-dried) powder. |
Biological Activity: | The activity is determined by the induced expression of CD25 on CD8+ mouse T cells which is typically detectable at concentrations starting at 0.1ng/ml. |
Reconstitution: | When reconstituting the product, gently pipet and wash down the sides of the vial to ensure full recovery of the protein into solution. It is recommended to reconstitute the lyophilized product with sterile water at a concentration of 0.1 mg/ml, which can be further diluted into other aqueous solutions. |
Shipping: | Ambient Temperature |
Long Term Storage: | -20°C |
Use/Stability: | It is recommended that a carrier protein (0.1% HSA or BSA) is added for long term storage. |
Handling: | Centrifuge the vial before opening the cap. After reconstitution, prepare aliquots and store at -20°C. |
Scientific Background: | Interleukin-12 (IL-12) is an important mediator of cellular-immunity produced by dendritic cells, macrophages and B cells when stimulated with antigen. IL-12 is involved in differentiating precursor helper T cells to Th1 and/or Th2 subtypes and promotes the production of IFN-γ and TNF-α. Mouse IL-12 is active on human and mouse cells. |

Regulatory Status: | RUO - Research Use Only |

Figure 2: 1µg in each lane (-) non-reducing conditions and (+) reducing conditions in a 4-20% TRIS-Glycine gel, stained with Coomassie Blue. Mouse IL-12 is a heterodimer comprised of one 40 kDa subunit and one 35 kDa subunit. The total predicted MW is 70 kDa.
Figure 1: Mouse IL-12 Bioactivity Data. Serial dilutions of mouse IL-12 (starting at 33ng/ml) were added to mouse splenic T cells. After 72 hours, cells were stained to analyze surface CD8 and CD25. The linear portion of the curve was us used to calculate the ED50. The ED50 for this lot of mouse IL-12 was between 10-20ng/ml.
