[K(Ac)40]α-Tubulin polyclonal antibody Western blot
Treatment of A549 cells with the histone deacetylase inhibitor trichostatin A (TSA, 0.3 µM, 7 hr.) induces acetylation of alpha-tubulin-Lys40. Western blot analysis was performed with A549 cell extracts (100,000 cells per lane) from either vehicle treated (Control) or TSAtreated (+TSA) cells. All lanes were probed with a 1:1000 dilution of the primary antibody, anti-alpha-tubulin (K(Ac)40) (Prod. No. BML-SA452). For lanes 2 and 5 the primary antibody was preincubated with the immunogen peptide (Ac-QMPSD-K(Ac)-TIGGG[CNH2] and for lanes 3 and 6 it was preincubated with non-acetylated peptide Ac-QMPSDKTIGGG[C-NH2] (peptides: 0.5 µg/µl, antibody: 1:100 dilution, 37°C, 1hr.). Secondary antibody for all lanes was donkey anti-rabbit-AP (1:2000; Jackson Immunoresearch) and the nitrocellulose membrane was developed with BCIP/NBT.
