Protocol:Immunohistochemistry on paraffin sections:
- Fix paraffin sections in 10% formalin.
- De-wax sections and block endogenous peroxide by incubating sections with 3% H2O2 for 15 min. at room temperature (RT).
- Wash sections with PBS and incubate with 2% normal rabbit serum at room temperature to block non-specific binding sites.
- Wash sections with PBS and incubate with primary antibody (anti-chemokine receptor) at recommended dilution for 60 min. at RT.
- Wash slides and incubate at RT with biotinylated rabbit anti-goat IgG H+L (Vector Labs) at 1:200 dilution for 30 min.
- Wash slides and incubate with HRP-Streptavidin (Vector Labs) 1:400 dilution for 30 min.
- Wash slides and incubate with DAB for 5-7 min. at RT.
- Wash slides with PBS and counter stain with hematoxylin.
