Western blot analysis of recombinant human sirtuins, Jurkat cells and HeLa nuclear extract. The nitrocellulose was probed with anti- SIRT1 at 1:1000. The secondary antibody was coupled to alkaline phosphatase and the blot was developed with BCIP/NBT. Arrows indicate positions of molecular weight markers. Lanes: S2, S3, S5 (SIRTs 2, 3 & 5, 50 ng each); J (150,000 Jurkat cells); H (18 μg HeLa nuclear extract); S1 (SIRT1- 5 ng, 0.5 ng).
