Serine Proteases as Cancer Biomarkers

Serine proteases (or serine endopeptidases) are enzymes that breakdown proteins in extracellular compartments by cleavage of peptide bonds in the proteins. There are at least 14 family members found ubiquitously in both eukaryotes and prokaryotes. In humans, they are responsible for coordinating various physiological functions including digestion, immune response, blood coagulation and reproduction. Dysfunction often leads to human diseases and disorders, and mutations may lead to decreased or increased enzymatic activity. In cancer, serine proteases mediate a variety of events relevant to fundamental processes of tumor invasion and metastasis. In ovarian cancer, they are thought to promote tumor growth and dissemination, and their production can be detected early in tumor formation. The link between serine protease functions and cancer outcomes suggests that they may be a suitable target for therapeutic agents aimed at prevention of tumor progression and metastasis. As a tumor biomarker, serine proteases hold promise as a means of identifying certain cancers at an earlier stage to guide treatment and improve outcome.
