Scientific Background: | Interferon-inducible T-cell α chemoattractant (ITAC), belonging to the CXC chemokine family, is expressed in peripheral blood leukocytes, pancreas and liver, thymus, spleen, lung, small intestine, placenta and prostate. Its molecular weight is 8–11 kDa. The cytokine is involved in regulating calcium release and inducing a chemotactic response in activated T-cells and is the dominant ligand for CXC receptor-3. The gene encoding this protein contains 4 exons and at least three polyadenylation signals which might reflect cell-specific regulation of expression. IFN-γ is a potent inducer of transcription of this gene. It is upregulated in patients with AIDS, dementia and in patients with multiple sclerosis. |

Western blot analysis of human ITAC was performed by loading 0.25 μg of recombinant human ITAC and induced HeLa cell culture supernatant onto a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked with 5% Milk/TBST for at least 1 hour. Membranes were probed with ITAC, mAb (22F1) (biotin conjugate) at a dilution of 1:1000 overnight at 4°C on a rocking platform. Membranes were washed in TBS-0.1%Tween 20 and probed with a goat anti-mouse-HRP secondary antibody. Membranes were washed and chemiluminescent detection performed.
