Scientific Background: | NEDD8 is a ubiquitin-like molecule, sharing ~60% identity with ubiquitin at the amino acid level, that can be covalently conjugated (neddylation) to a limited number of cellular proteins in a fashion similar to that of ubiquitin. The C-terminus of pro-NEDD8, GGLRQ, is efficiently processed to expose glycine76 in the mature form. This processing is required for conjugation to target proteins via a range of specific activating and conjugating enzymes (Gong and Yeh, 1999). The E1 activating-enzyme complex is composed of two subunits, APP-BP1 and UBA3, whilst the E2 conjugating-enzyme is the ubiquitin E2 UbcH12. The expression of NEDD8 is essential for growth in most organisms where it has been shown to modify the cullin subunit of cullin-based ubiquitin-protein ligases (E3s). Neddylation acts to regulate the function of these E3s and organisms with lesions in the neddylation process exhibit severe growth defects (Parry and Estelle, 2004). Previously, it was believed that the cullins were the only substrates for NEDDylation, however, it is now recognised that there are a number of such proteins, including the von Hippel-Lindau (VHL) tumour suppressor gene (Stickle et al., 2004) and the tumour suppressor and transcriptional regulator p53 (Harper, 2004) amongst others. |
