Scientific Background:MKP-1 is the founding exemplar of a group of dual specificity phosphatases which negatively regulate the MAP Kinases, dephosphorylating both the phosphotyrosine and the phosphothreonine in the -pTXpY- activation-loop motif. MKP-1 is implicated primarily in the inactivation of the stress-responsive p38 and JNK MAP kinase. Studies indicate that MKP-1 is the anti-inflammatory target of glucocorticoids in human endothelium.
