Scientific Background: | The melanocortin receptors are members of the G protein-coupled receptor superfamily. Their ligands, the melanocortins, are peptides consisting of µ-, β- and γ-melanocyte stimulating hormone and adrenocorticotropic hormone. MC3R binds all melanocortins with equal affinity. Agouti-related protein (AGRP) is the high affinity endogenous antagonist of MC3R. It is expressed in the brain, placenta, and gut tissues, but not in the adrenal cortex or melanocytes. |

Technical Info/Product Notes: | PREADSORPTION CONTROL: 1µg peptide per 1µg antibody. Prepare 2 tubes. In the first, dilute the necessary amount of antibody in 200-500µl of PBS, 1% BSA and 0.025% sodium aide. Prepare the second tube identically to the first with the addition of the appropriate amount of peptide. Incubate at room temperature for 1 hr. Centrifuge both tubes at 10,000Xg for 5 min. Use the supernatants from tubes 1 and 2 for parallel experiments. |
