Scientific Background:When cells are stimulated with pro-inflammatory cytokines, most of their constitutively expressed proteasomes are replaced with immunoproteasomes, which increase the production of peptides for presentation on MHC class I molecules. In addition, cortical thymic epithelial cells selectively express a type of proteasome known as the thymoproteasome, in which the β5i immunoproteasome subunit has been replaced by the thymus specific proteasome subunit β5t. The thymoproteasome is required for the positive selection of thymocytes and shaping the T cell receptor repertoire of cytotoxic T lymphocytes. In addition mice deficient in β5t display defective development of CD8(+) T cells in the thymus suggesting a key role for β5t and the thymoproteasome in generating MHC class I-restricted CD8(+) T cells.
 
Technical Info/Product Notes:The hybridoma secreting the antibody clone b8-17 was generated by fusion of splenocytes from Balb/cmice which had received repeated immunisation with recombinant human β5t subunit protein (accession number: A5LHX3). Immunoglobulin was purified from tissue culture supernatant by ion exchange chromatography.
