Scientific Background: | The 19S regulatory complex consists of a ‘base’ sub-complex, composed of a hetero-hexameric ring of AAA-ATPases that not only function as the gate-opener to the 20S proteasome but also, in conjunction with two other high-molecular-mass non-ATPase subunits (Rpn2/S1 and Rpn1/S2), as a platform for protein unfolding. The ‘lid’ sub-complex of the 19S complex, composed of non-ATPase subunits is involved in the binding and deubiquitinylation of proteasome targeted ubiquitin conjugated proteins and assists in the transport of substrates through the ‘base’ and into the 20S catalytic core. |
