A novel small molecule NGF mimic discovered using a cell-based chemical genetic screen for TrkA agonists. Gambogic amide selectively binds to TrkA, triggers its tyrosine phosphorylation, elicits PI3-kinase/Akt and MAPK activation, provokes neurite outgrowth in PC12 cells and prevents neuronal cell death. EC50 for antiapoptotic activity is 5nM (see reference S.-W. Jang, et al. 2007).
