Scientific Background:IL-33 gene was initially designated as NF-HEV which encodes a predicted 270-residue protein. The calculated 30 KDa human IL33 is converted by CASP1 to a mature 18-KD
protein. IL-33 binds to and signals through ST2 (IL1R1) and its stimulation recruits MYD88, IRAK, IRAK4, and TRAF6, followed by phosphorylation of ERK1(MAPK3)/ERK2(MAPK1), p38(MAPK14), and JNK.
Nuclear localization, heterochromatinassociation, and targeting to mitotic chromosomes were all found to be mediated by an evolutionarily conserved homeodomain-like helix-turn-helix motif within the IL-33 N-terminal part. IL-33 was found to possess transcriptional repressor properties, associated with the homeodomain-like helix-turn-helix motif.
