Scientific Background: | Beta-amyloid/A-beta is the major protein component of neurofibrillary tangles and senile plaques, major neruopthological hallmarks of Alzheimer's disease (AD). Abeta occurs in two predominant forms with different COOH-termini, A-beta 40 and A-beta 42. A-beta is generated by proteolytic cleavage of the amyloid precursor protein (APP) at two sites by distinct secretases, the beta-site secretase BACE, and the multi-subunit gamma-secretase composed of presenilin, nicastrin, Aph-1, and Pen-2. |
