Scientific Background: | Substance P (SP) is an 11 amino acid member of the tachykinin family of neuropeptides which includes SP, neurokinin-A (NK-A) and neurokinin-B (NK-B), all of which exhibit a common COOH-terminal pentapeptide sequence Phe-X-Gly-Leu-Met-NH2. SP functions as a neurotransmitter and its effects are associated with physiological processes such as vomiting, pain, and vasodilation. Three tachykinin receptors have been identified to date, NK1, NK2, and NK3, with SP showing preferential affinity for the NK1 receptor. SP signaling occurs primarily via coupling of Gq/11 alpha subunits to NK1, leading to phospholipase C activation. High SP concentrations may also stimulate adenylyl cyclase activity, resulting in elevated cAMP. |

Western blot analysis: MW marker (1) and 25µg rat brain extract (2) probed with Substance P Receptor (NK1) pAb at 3.7µg/ml
Membrane ELISA: Lewis rat striatum membranes (5µg/well) were treated with 1 μM concentrations of agonist (Substance P) and probed with Substance P Receptor (NK1) pAb (1:250 to 1:2000 of a 1µg/μl stock solution) by ELISA. Data from vehicle treated cells were taken as 100%. Results are the mean ± SEM (n=4).
