Scientific Background: | Bcl-2 (B-cell lymphoma 2) is an oncogenic protein that inhibits apoptosis. It is located within the mitochondrial membrane, endoplasmic reticulum and nuclear envelope. Bcl-2 is encoded by a 230kb gene that results in a 24-26kDa protein4. Bakhshi first identified Bcl-2 as an apoptotic proto-oncogene while studying chromosomal translocations in human follicular lymphomas. At approximately the same time Tsujimoto and Croce made the same observation. The function of Bcl-2 was first described by Vaux when it was demonstrated that it promoted the survival of factor dependent cells. Bcl-2 overexpression has been implicated in a wide variety of malignancies including (but not limited to) breast, prostate, skin, colon and pancreatic cancers. Related anti-apoptotic proteins include Bcl-xL, Mcl-1 and Bcl-w. Pro-apoptotic proteins in the Bcl-2 family include Bax, BAD, Bak, Blk and Bid. |
