Selective degradation of intracellular targets, such as misfolded proteins and damaged organelles is an important homeostatic function of the cell. Misfolded proteins, as well as, aged or defective organelles that occur with day-to-day stress are eliminated by autophagy (literally meaning eating oneself).
Under physiological conditions, autophagy plays a variety of important roles including maintenance of the amino acid pool during starvation, prevention of neurodegeneration, tumor suppression, cellular differentiation, clearance of intracellular microbes and regulation of innate and adaptive immunity. Autophagy is considered a dynamic, multi-step process which can be regulated at several steps, in both a positive and negative manner.
Cyto-ID™ Autophagy detection dye becomes brightly fluorescent in vesicles produced during conditions known to modulate the autophagy pathway. The probe does not respond to apoptotic signals.
