The peptide consists of two domains - an antennapedia homeodomain that confers cell permeability and the T735 to E745 region of IKKβ which is the NEMO binding domain (NBD). It is a selective inhibitor of NFκB activation. Blocks interaction of NEMO (an IKB complex regulatory protein) with the IκB kinase complex. The peptide (100 µM) inhibits E-selectin expression in human umbilical vein endothelial cells treated with TNFα and inhibits LPS-induced nitric acid release from a macrophage cell line In an experimental mouse model of acute inflammation, IKK-NBD peptide (topically applied) inhibited PMA induced ear inflammation.
