A QUANTIZYME® Assay System. The Cathepsin S Drug Discovery Kit is a complete assay system designed to screen inhibitors of cathepsin S. Using a convenient microplate format, cathepsin S activity is monitored fluorimetrically using the substrate provided in the kit.
Kinetics of Z-VVR-AMC cleavage by Cathepsin S, 0.059mU/well; 37°C. Rates were obtained from the slope of the initial, linear portion of plots of AFU vs. time. Curve and kinetic parameters derived from a non-linear least squares fit to the Michaelis-Menten equation (Marquadt algorithm).
Example of inhibitor data. Using the control, the slope is equal to 34.28 RFU/sec. With the inhibitor (E-64), the slope is equal to 0 RFU/sec. The % activity remaining after using the inhibitor can be calculated as follow: (0/34.28) x 100 = 0%

Use/Stability: | The enzyme component should be thawed as close to the time of use as is possible, and held on ice while in use. The enzyme can be frozen and thawed up to 5 times, but care should be taken to minimize both the time the enzyme spends on ice, and the number of times it is frozen and thawed. Do not hold the diluted enzyme at assay temperature (e.g 37°C) for an extended time before starting the assay. |
