The YVAD-CHO portion of this peptide is a highly-specific, potent (Ki=1 nM) and reversible inhibitor of caspase-1 (interleukin-1β converting enzyme, ICE), whose sequence is based on the pre-interleukin-1β Asp-116 cleavage site for caspase-1. The sequence linked N-terminal to YVAD-CHO (residues 1-16) has been shown to enhance cell permeability. The efficacy of the resulting chimeric peptide, for caspase-1 inhibition, is unknown.

Use/Stability: | Preincubate cultured cells 1-2 hrs. with 0.1-2.0µM of the peptide, before inducing for apoptosis. Measure indicator(s) of apoptosis at a number of time points, since the effect of the inhibitor may be a delay in the onset of apoptosis, rather than permanent inhibition. It may also be helpful to determine a minimum effective concentration for the apoptosis inducer and use that concentration, when attempting to observe the effects of this peptide. |
