Scientific Background: | Decoy receptors (DcRs) are signaling-deficient receptors for certain members of the TNF family of cytokines, including TRAIL/Apo2L and FasL. Functional TRAIL receptors such as DR4 and DR5 transmit apoptotic signals via their intracellular death domains. In contrast, three identified TRAIL decoy receptors (DcR1, DcR2, and osteoprotegerin/OPG) completely or partially lack the signaling death domain, and/or are secreted from cells and unable to transmit intracellular signals. Thus, expression of DcRs promotes survival by sequestration of apoptotic ligands. A fourth identified decoy receptor, DcR3, binds both FasL and the vascular endothelial cell growth inhibitor (VEGI), inhibiting Fas and DR3 death signals. |
