Assay Kits, Enzymes, and Substrates for HTS

Enzyme Activity Assays

Z’-Factor determination for the Fluor de Lys®-Green HDAC2 assay

An extensive range of assay kits, enzymes, and substrates is available to assist with primary and secondary screening. HTS-friendly, off-the-shelf assays are supplied with enzyme, substrate and detection reagents ready for straightforward evaluation by your assay development team. Active, quality-controlled enzymes and sensitive substrates are available for important drug discovery targets including:

FluoForte™ Calcium Assays for GPCR & Calcium Channel Screening

FluoForte™ generates the highest intensity signal when compared to Fluo-4 Direct™ and Calcium 4.

Based upon a new fluorescent calcium indicator, FluoForte™ calcium assay kits offer superior calcium mobilization assays for screening GPCRs and calcium channels. FluoForte™ is more than 2-fold brighter than Fluo-4, yields more robust signal (Z’-factor ≥ 0.75) and provides a larger assay window; making it suitable for challenging cell lines or receptors and increasing the probability of detecting weak agonists.

  • Homogeneous mix-and-read, no-wash calcium mobilization assay
  • Load dye in live cells at 37°C or even room temperature for easier automation
  • Provides comparable EC50 values to commercially available calcium assays
  • No quencher dye - no interference from compound ligand-receptor interactions

ProteoStat™ Thermal Shift Assay

The ligand TFMSA binds to Carbonic Anhydrase I and stabilizes the protein’s structure.

Determining the optimal conditions that prevent aggregation of protein targets during storage and handling is critical to a successful high-throughput screen. In addition, determining that hits from screening are due to inhibition and not aggregation effects is an important secondary screen in the hit-to-lead process.  The ProteoStat™ Thermal Shift Stability Assay is a rapid, simple fluorescent screening method that detects protein aggregates.

  • Determines the impact of ligand-protein interactions on target protein stability
  • Accelerated analysis of parameters impacting monoclonal antibody and recombinant protein stability
  • Measures aggregation temperature
