Scientific Background: | IFN-α-2 is a cytokine that displays antiviral and antiproliferative properties. It is comprised of three major subtypes, which include IFN-α-2a, IFN-α-2b, and IFN-α-2c. IFN-α-2b is the predominant subtype in the human genome. It binds to its receptor and activates the JAK-STAT pathway, which stimulates expression of the host genes responsible for antiviral activity. IFN-α-2b is used to treat many diseases, such as hepatitis B and C, melanoma, Kaposi's sarcoma, chronic myeloid lymphoma, and angioblastoma. The combination of IFN-α-2b with ribavirin, a viral inhibitor, has become the standard treatment for chronic hepatitis C virus infection. |

Coomassie stained SDS-PAGE. Lane 1, Molecular weight marker. Lane 2, 1.0µg Human IFN-α-2b (ENZ-PRT192).
ENZ-PRT192 activity was determined by comparing against the WHO International Standard for Interferon Alpha 2b (NIBSC, Code 95/566) using the HEK-Blue™ hTLR5 reporter cell line (Invivogen). Briefly, cells were plated at 50,000 cells per well in 100μl of test media. Known amounts of WHO standard and serially diluted ENZ-PRT192 were added in a volume of 100μl. The cells were grown for 20-24 hours at 37°C + 5% CO2. After incubation, 180μl of Quanti-Blue™ was added to 20μl of HEK-Blue™ hTLR5 reporter cell supernatant and the color was allowed to develop for 3 hours at 37°C + 5% CO2. Activity was determined by interpolating the OD at 650nm of the ENZ-PRT192 titer off of the WHO standard curve.
