Scientific Background: | The HER2/Neu (c-erbB-2) proto-oncogene is a trans-membrane receptor tyrosine kinase that is clinically indicated in a number of carcinomas. Overexpression of the HER2/Neu protein has been associated with ductal breast cancer, as well as pulmonary and gastric adenocarcinomas. A correlation between HER2/Neu and p53 has also been documented, as overexpression of both proteins has been associated with early invasion and metastasis in bladder cancer. |

Protocol: | Manual Use
- Antigen Retrieval: Perform antigen retrieval with Antigen Retrieval Reagent, pH 9 (ENZ-ACC113), for 10 to 30 minutes at 100°C.
- Peroxidase Block: Block using IHC/ISH Peroxidase Block (ENZ-ACC107) for five minutes at room temperature.
- Primary Antibody: Dilute antibody 1:100-1:200 before applying 150 µL directly to the tissue. Incubate for 10 to 30 minutes at room temperature.
- Secondary Antibody: Incubate using POLYVIEW® PLUS (anti-Mouse) HRP Reagent (ENZ-ACC104) or POLYVIEW® PLUS (anti-Mouse) AP Reagent (ENZ-ACC114) for 20 to 30 minutes.
- Substrate Development: Incubate using HIGHDEF® DAB Substrate/Chromogen (ENZ-ACC105) for 5 to 10 minutes at room temperature.
- Counterstain: Counterstain with hematoxylin for 30 seconds to 5 minutes. Rinse with distilled water.
Automated Staining
For automated staining, we recommend incubating for 30 minutes with the primary antibody and using Antigen Retrieval at pH 9 for 30 minutes. |
