Scientific Background:Deposition of APP (Amyloid A4 protein) precursor intraneuronally as neurofibrillary tangles, extracellularly as plaques, and in blood vessels, is characteristic of both Alzheimer's disease and aged Down's Syndrome. There are five forms of APP: APP395, APP563, APP695, APP751, and APP770. They are produced by alternative splicing of the APP gene. APP695 is the most predominant form in neuronal tissue while APP751 is the predominant form found elsewhere.

HEK cells were transfected with pCDNA3-APP695 (lane 2), pCDNA3-APP695 ΔC31 (lane 3) or pCDNA3 (lane 4) constructs. 10µg of total cell lysates were separated by 7.5% SDS-PAGE for Western Blot. The membrane was immunoblotted with primary antibody (ENZ-ABS226, APP, pAb) at 1:1000 dilution. Anti-rabbit IgG-AP (Prod. No. ADI-SAB-301) was used at 1:4000 dilution. The result showed this antibody detected both full length APP695 and APP695 ΔC31 at about 98 KDa. Pre-stained protein marker is shown in lane 1.
