BioArray™ cDNA Synthesis Kit, is based on the scheme of Van Gelder et al. (1990). An oligo dT primer with a bacteriophage T7 promoter sequence at its 5’-end is hybridized to poly A sequences in mRNA. First strand cDNA synthesis proceeds from the action of reverse transcriptase. Second strand synthesis is accomplished by the combined activity of RNase H, to create nicks in the RNA/DNA hybrid, and DNA polymerase that uses these nicks as priming sites for second strand cDNA synthesis.
