Miscellaneous/General:Vps9p is a Saccharomyces cerevisiae exchange factor for the yeast Rab5 GTPase homolog Vps21p, and is known to promote mono-ubiquitylation. Vps9p contains a CUE domain, which shares structural similarity to ubiquitin-associated (UBA) domains. The CUE domain of the yeast Cue2 protein binds a region of ubiquitin that includes the hydrophobic patch , Leu8-Ile44-Val70, and encompasses Lys48, a site of polyubiquitin chain formation. Studies have demonstrated that, in addition to binding mono-ubiquitin, an isolated CUE domain can bind multi-ubiquitin chains and ubiquitylated substrates.
