Scientific Background: | UbcH8 is a class I ubiquitin-conjugating (E2) enzyme highly similar in structure to UbcH7. UbcH8 has been demonstrated to act in conjunction with Parkin, a ring-finger-containing E3 ubiquitin protein ligase implicated in the pathogenesis of autosomal recessive familial Parkinson's Disease, and with Staring, an ring-finger E3 ligase responsible for ubiquitinylation of syntaxin 1. UbcH8 is a primary target gene of interferon-α and γ, and recent studies have indicated that UbcH8 is the E2 enzyme for the ubiquitin-like protein ISG15 (also known as UCRP). These studies suggest overlap between the ISG15 and ubiquitin conjugation pathways, and have important implications for understanding the regulation and function of ISG15 in the IFN α/β response. |
