Scientific Background:In addition to binding TGF-β and β-amyloid, α2-macroglobulin covalently binds and inhibits a wide range of proteases, including ADAMs, calpains, cathepsins, chymase, clotting factors, kallikreins, MMPs, neutrophil elastase, and proteinase 3.  It also plays a role in the innate immune system, inhibiting parasitic proteases.  When treated with methylamine, it blocks binding of apoE to lipoprotein receptor-related protein (LRP).
