| Miscellaneous/General: | USP18 (UBP43) is a specific deconjugating enzyme for ISG15, a vertebrate ubiquitin like protein strongly upregulated by type I interferon following bacterial or viral infection. Whilst the molecular and regulatory consequences of ISGylation are not fully understood, the absence of USP18 has a profound effect on innate immunity, leading to increased resistance to certain viral infections. Notably, these effects do not appear to be contingent upon the proteolytic activity of USP18 suggesting additional roles for USP18 over and above that of an ISG15 specific protease. USP18 has been shown to significantly limit operation of the extrinsic apoptotic pathway triggered by type I interferon and drugs. |
