For detection of tripeptidyl peptidase II (TPPII). TPPII plays a critical role in cleaving proteasomal-produced peptides into tripeptides that can be further degraded by other peptidases. In addition to a physiological rôle that is concerned with general protein turnover, there are more specific rôles for TPPII, such as its rôle in major histocompatibility complex (MHC) class I antigen processing.

Scientific Background:TPPII plays a critical role in cleaving proteasomeal-produced peptides into tripeptides that can be further degraded by dipeptidyl peptidase II (DPPII) and aminopeptidases. In addition to a physiological rôle that is concerned with general protein turnover, there are more specific rôles for TPPII, such as its rôle in major histocompatibility complex (MHC) class I antigen processing in the cytosolic subtilisin. In addtion to exo-peptidase activity, TPPII also exhibits endo-proteolytic cleavage properties. TPPII may also generate antigenic peptides for the transporter associated with antigen processing (TAP) and appears to be an important intermediate between the proteasome and the rest of the peptidase pool involved in trimming of larger fragments to antigenic peptides and mainly free amino acids.
