The assembly of mammalian 20S proteasomes is an ordered multi-step process, starting from a-ring formation, that is in part mediated by the proteasome-specific chaperones PAC1 and PAC2. A third human 20S proteasome chaperone PAC3 has been identified as a component of the a-ring-PAC1-PAC2 complex. PAC3 is proposed to co-operate with PAC1-PAC2 heterodimers to orchestrate the correct assembly of 20S proteasomes.
