The FLUOR DE LYS®-Succinyl, Desuccinylase Substrate offers a highly sensitive and convenient alternative to radiolabeled, succinylated histones or peptide/HPLC methods for the assay of desuccinylases such as SIRT5. The assay procedure has two steps. First, the FLUOR DE LYS®- Succinyl, Desuccinylase Substrate, which comprises a succinylated lysine side chain, is incubated with a sample containing desuccinylase activity (HeLa nuclear or other extract, purified enzyme, bead-bound immunocomplex, etc.). Desuccinylation of the substrate sensitizes the substrate so that, in the second step, treatment with the FLUOR DE LYS® Developer (Prod. No. BML-KI105) produces a free AMC fluorophore which can be monitored with standard fluorometers (Ex. 350-380nm / Em. 450-480nm).
