Fluor de Lys® Developer is an essential reagent for signal generation in HDAC or sirtuin assays done with the Fluor de Lys
® substrate. An appropriate Fluor de Lys
® family substrate, comprising an acetylated lysine side chain, is first incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes it so that, in a second step, treatment with the Fluor de Lys
® Developer produces a fluorophore.
Typically, an ‘inhibitor stop’ will be included in the 1x Developer dilution (e.g. Trichostatin A) for class I and II HDACs, nicotinamide for sirtuins).
