ALX-804-401-C100 100 µg 451.00 USD

Product Specification

Alternative Name:Protein kinase B, PKB

Purity Detail:Purified from serum-free cell culture supernatant by subsequent thiophilic adsorption and size exclusion chromatography.

Formulation:Lyophilized from 1ml 2x PBS containing 0.09% sodium azide, PEG and sucrose.

Clone:11E6

Isotype:Mouse IgG1

Immunogen:Synthetic peptide corresponding to C-terminal (PQFpSYSA) of human Akt (protein kinase B; PKB) phosphorylated at Ser473.

Reconstitution:Reconstitute with 1ml distilled water (15 min., RT).

Specificity:Recognizes human, mouse, rat and dog Ser472/473/474-phosphorylated Akt (see technical note). Does not cross-react with dephosphorylated Akt or other unrelated serine-phosphorylated proteins.

Application:Flow Cytometry: (1μg/μm)
Immunohistochemistry: Optimal conditions must be determined individually. A starting dilution of 5μg/ml is recommended.
Western Blot: (0.5µg/ml for HRPO/ECL detection).
Recommended blocking buffer CPPT: 10mM TRIS-HCl, pH 7.4, 0.5% (w/v) casein, 1% (w/v) PEG 4’000, 1% (w/v) polyvinylpyrrolidone, 0.1% (v/v) Tween 20, 150mM sodium chloride.
Optimal conditions must be determined individually for each application.

Long Term Storage:-20°C

Use/Stability:

Reconstituted antibody is stable for 1 year when stored at -80°C. Thaw aliquots at +37°C. Thawed aliquots may be stored at +4°C for up to 3 months.

Handling:After reconstitution, prepare aliquots and freeze in liquid nitrogen.
Avoid freeze/thaw cycles.

Positive Control:Included (Prod. No. ALX-840-025).

Background / Technical Information:Technical Note: There are three mammalian isoforms of Akt: Akt1 (PKBa), Akt2 (PKBb), and Akt3 (PKBg). Akt1 shares 81% and 83% amino acid identity with Akt2 and Akt3, respectively. Although the three isoforms show broad tissue distribution, Akt1 is the most ubiquitously expressed. Akt2 is expressed at a lower level than Akt1 except in insulin-responsive tissues where it predominates. In particular, Akt2 is more abundant and more highly activated than Akt1 in adipocytes. Akt3 is expressed at the lowest level except in testes and brain. The C-terminal sequence of the three isoforms are shown below. Note that the phosphorylation site slightly differs between the three isoforms. Thus the phosphorylation site Ser473 of Akt1 corresponds to Ser474 in Akt2 and Ser472 in Akt3 respectively.

Akt2: L461LELDQRTHFPQFS474YSASIRE481 UniProt ID P31751
Akt3: D461NERRPHFPQFS472YSASGRE479 UniProt IDQ9Y243
