A20 is a zinc finger protein that renders cells resistant to apoptosis. A20-deficient mice develop severe inflammation and are hyper-responsive to LPS. S.M. O'Reilly & P.N. Moynagh have shown that full length A20 inhibits activation of NF-kB and AP-1 by TLR4. A20 associates with A20-binding inhibitor of NF-kB (ABIN)-1 and -2, which interfere with NF-kB activation upstream of the IKK complex.

Method: HT1080 cells, wild type or transfected with human A20 were activated with 0.1µg/ml of rhsTNF-a (Prod. No. ALX-522-008) for 4 hours. Cells were then harvested, lysed and the cell extracts were resolved by SDS-PAGE under reducing conditions, transferred to nitrocellulose and incubated with the 8E8.38 monoclonal antibody (1:1000 dilution O/N). Proteins were visualized using a peroxidase-conjugated antibody to mouse IgG1 and a chemiluminescence detection system.
