Scientific Background:The fibroblast growth factor-inducible-14 (Fn14) is a 129 aa type Ia transmembrane protein. Fn14 is a growth-factor regulated, immediate-early response protein. It is expressed in a developmental and adult tissue-specific manner. Fn14 expression decreases cellular adhesion to extracellular matrix proteins fibronectin and vitronectin and reduces cell growth and migration. It is expressed at high levels in liver cancer cells, in hepatocellular carcinoma specimens as well as in regenerating liver cells showing Fn14 may play a role in hepatocyte growth and liver neoplasia. Fn14 has been described to bind TWEAK (Apo-3 Ligand), a member of the TNF family. Historical data has shown that Fn14 inhibits TWEAK-mediated killing of Kym-1 target cells.
Figure: HEK 293T cells were mock transfected (thin line) or transfected with an expression plasmid coding for the extracellular domain of human TWEAK (thick line).
Method: Cells (5 x 105) were incubated on ice for 30 min in 50µl FACS buffer (PBS, 5% Fetal calf serum, 0.02% azide) containing 1µg/ml of mFn14:Fc-FITC. After washing cells were analyzed by flow cytometry.
