Protocol:Flow Cytometry Protocol:
- Wash cells in PBS, and resuspend in binding buffer (10mM HEPES/NaOH, pH 7.4, 140mM NaCl, 2.5mM CaCl2; filtered through 0.2µm filter).
- Adjust cells to a density between 200’000 and 500’000 cells/ml.
- Take 195µl cell suspension and add 5µl Annexin-ATTO 647N. Mix and incubate 10 min. in the dark.
- Wash cells 1x and resuspend in 190µl binding buffer.
- Add 10µl of 20µg/ml propidium iodide (PI) stock solution to stain for necrotic cells.
- Analyze by FACS.
Note: These are general recommendations. Optimal conditions must be determined individually for each application.
