| Background / Technical Information: | Flow Cytometry Protocol:- Wash cells in PBS, and resuspend in binding buffer (10mM HEPES/NaOH, pH 7.4, 140mM NaCl, 2.5mM CaCl2; filtered through 0.2µm filter).- Adjust cells to a density between 200’000 and 500’000 cells/ml.- Take 195µl cell suspension and add 5µl Annexin-ATTO 488. Mix and incubate 10 min. in the dark.- Wash cells 1x and resuspend in 190µl binding buffer.- Add 10µl of 20µg/ml propidium iodide (PI) stock solution to stain for necrotic cells.- Analyze by FACS.Note: These are general recommendations.
