No. ALX-201-356): Hydrolysis of the fluorescent peptide substrate Mca-YVADAPK(Dnp)-OH.Method: To each test tube, 2ml of assay buffer (75mM TRIS, pH 7.4, 1M sodium chloride, 10µM zinc chloride, 10µM fluorescent peptide substrate) was added. Reactions were initiated by addition of 20µl (1µg/ml, final concentration 10ng/ml) of ACE2, Soluble (human) (recombinant) (FLAG®), a FLAG tag control protein and buffer only (for blank). Reactions were performed for 20 min. at room temperature and stopped by addition of 0.2ml of 500mM EDTA. Fluorescence intensity was measured using luminescence spectrometer (Ex=320nm, Em=405nm).
