| Miscellaneous/General: | SAE1/SAE2 is a heterodimer that acts as the activating enzyme for SUMO-1, SUMO-2 and SUMO-3 and catalyzes formation of a thioester bond between the C-terminus of SUMO/sentrin (aa 1-347) and a cysteine residue in SAE2 (aa 1-640). In the presence of Ubc9, SAE1/SAE2 transfers the activated SUMO to a cysteine residue in Ubc9. |

| Background / Technical Information: | Standard reaction for protein conjugation (10µl) includes the target protein, SUMO-1 activating enzyme (Prod. No. ALX-201-090) (100ng), UbcH9 (Prod. No. ALX-201-046) (900ng), SUMO (10µg) in 50mM TRIS, pH 7.6, 5mM magnesium chloride, 2mM ATP, 10mM creatine phosphate, 3.5U/ml of creatine kinase and 0.6U/ml of inorganic pyrophosphatase. 10µg are sufficient for about 100 assays.
