Scientific Background:The p90 Ribosomal S6 kinases (RSKs) comprise a family of unique protein serine/threonine kinases that lie downstream of the mitogen activated protein kinases, predominantly ERK. Activation of ERK results in the phosphorylation and subsequent activation of RSKs. In turn, RSKs translocate to the nucleus where they phosphorylate a diverse range of nuclear substrates including c-Fos, estrogen receptor, NF-κB/IkappaBalpha, cAMP-response element-binding protein (CREB) and CREB-binding protein. RSKs play a critical role in mediating a number of biological responses including cell cycle regulation and protein synthesis.
