ADI-905-797-100 100 µg 270.00 USD

Product Specification

Purity Detail:Peptide affinity purified.

Formulation:Liquid. In PBS containing 50% glycerol and 0.01% sodium azide.

Immunogen:Synthetic peptide corresponding to the sequence near the N-terminus of rat muscarine receptor M2.

Source/Host:From rabbit.

Specificity:Recognizes rat muscarine receptor M2. Predicted MW of ~51kDa. Higher MW species (~150kDa and ~70kDa) also observed by Western blot which may reflect post-translational modification and heterodimerization of the receptor.

Application:ELISA (Membrane)
Western Blot
Optimal conditions must be determined individually for each application.

Long Term Storage:-20°C

Miscellaneous/General:The pharmacologic activity of acetylcholine (ACTH) is mediated by the ionotropic nicotinic receptor or via metabotropic muscarinic receptors. Five known acetylcholine (muscarinic) receptors (M1-M5) are variably expressed in the central nervous system and periphery, mediating motor control, thermoregulation, memory, smooth muscle contraction, glandular secretion, and cardiac function. Although the physiology of the five mammalian muscarinic receptor homologs does not differ significantly in their affinities to a variety of agonists (ACTH, muscarine) and antagonists (e.g., atropine), they can be divided into two sub-groups (M1/M3/M5 and M2/M4) based on G alpha subunit coupling preferences. M1/M3/M5 receptors signal primarily via Gq/11 alpha subunits to stimulate phospholipase C, whereas M2/M4 receptors couple to Gi/o alpha subunits to inhibit adenylyl cyclases and cAMP production. Modulation of muscarinic receptor activity holds promise for the treatment of psychiatric and neurological disorders including schizophrenia and Alzheimer's disease.

Manufacturer:Manufactured by Proteimax.

905-797 WB
Western blot analysis: MW marker (1) and 20µg rat heart extract (2) probed with Muscarine Receptor (M2) pAb at 4.2µg/ml.
905-797 m ELISA
Membrane ELISA: Lewis rat heart membranes (5µg/well) were treated with 1 μM concentrations of agonist (Bethanechol) and probed with Muscarine Receptor (M2) pAb (1:10 to 1:150 of a 0.1µg/μL stock solution) by ELISA. Data from vehicle treated cells were taken as 100%. Results are the mean ± SEM (n=3).
