Immunogen:Synthetic peptide corresponding to the sequence near the N-terminus of rat bradykinin receptor B2.

Source/Host:From rabbit.

Specificity:Recognizes mouse and rat bradykinin receptor B2. Predicted MW of ~45kDa. Higher MW species (~50kDa) and intermediates also observed by Western blot which reflect post-translational modification or dimerization of the receptor.

Application:ELISA (Membrane)
Immunohistochemistry
Western Blot
Optimal conditions must be determined individually for each application.

Long Term Storage:-20°C

Miscellaneous/General:Bradykinin (BK) is a nine-amino acid peptide produced by kallikrein cleavage of kininogen precursor proteins in tissue and plasma, and is an important regulator of vascular and pain responses to tissue injury. Two receptors (B1 and B2) have been identified as mediators of BK signaling, and are both seven-transmembrane domain-containing GPCRs. The B2 receptor is constitutively expressed in smooth muscle and neurons, whereas B1 expression is induced following injury. B2 receptors signal via Gq alpha subunits, leading to activation of phospholipase Cbeta, hydrolysis of PI, and an intracellular increase in free calcium, although coupling through Gi, Gs, and G12/13 has also been observed.

Manufacturer:Manufactured by Proteimax.

905-747 WB
Western blot analysis: MW marker (1) and 20µg rat brain extract (2) probed with Bradykinin Receptor (B2) pAb at 2.2µg/ml.
905-747 IHC
Immunohistochemistry analysis: Mouse brain fixed with 4% paraformaldehyde, stained with Bradykinin Receptor (B2) pAb at 1:1000, and visualized at 40x.
905-747 m ELISA
Membrane ELISA: Lewis rat heart membranes (5µg/well) were treated with 1 μM concentrations of agonist (BK) and probed with Bradykinin Receptor (B2) pAb (1:250 to 1:2000 of a 1µg/μL stock solution) by ELISA. Data from vehicle treated cells were taken as 100%. Results are the mean ± SEM (n=3).
